Principle of the Assay::The Human Biglycan ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Biglycan in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human Biglycan coated on a 96-well plate. Standards and samples are pipetted into the wells and Biglycan present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Biglycan antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Biglycan bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Biglycan (BGN) is a small leucine-rich repeat proteoglycan (SLRP) which is found in a variety of extracellular matrix tissues, including bone, cartilage and tendon. It is an important structural component of articular cartilage and participates in the assembly of the chondrocyte extracellular matrix through formation of protein interactions with type VI collagen and large proteoglycan aggregates. In humans, Biglycan is encoded by the BGN gene. It is mapped to Xq28. Biglycan is believed to play a role in the mineralisation of bone, and its core protein binds to the growth factors BMP-4 and influences its bioactivity. It has been reported that the presence of biglycan is necessary for BMP-4 to exert its effects on osteoblasts. There is also evidence that biglycan can bind to TGF-beta 1.
